In vitro characterization of the Gd complex of [2,6-pyridinediylbis(methylene nitrilo)] tetraacetic acid (PMN-tetraacetic acid) and of its Eu analogue, suitable bimodal contrast agents for MRI and optical imaging
[en] Gd and Eu complexes of PMN-tetraacetic acid show interesting properties either for MRI or for optical imaging; that is, for the Gd-complex, a high proton relaxivity with favorable water residence time; for the Eu-complex, a luminescence lifetime of 400 mu s at room temperature compatible with the use of time-resolved luminescence technique. Both complexes have a good stability in physiological medium.

The O-17 chemical shifts were measured at 332 K and 7.05 T (Bruker AMX-300 spectrometer) on solutions containing 15 to 20 mM of the Gd-complex dissolved in 1.7 ml of water added with 0.3 ml of D2O for the lock.

The overall luminescence quantum yield of the complex (Φtot) can be broken into two contributions: Φtot = ηsens × ΦEu, where ηsens is the efficiency of the sensitization of the Eu3+ ion through the ligand and ΦEu is the intrinsic quantum yield of the Eu3+ ion when it is excited in its own levels. The value of Φtot was determined experimentally by using [Ru(bpy)3]2+ in aerated water (Φ = 0.028) as reference. The value of ΦEu (0.09) was determined following the procedure described by Verhoeven et al. Werts, M. H. V.; Jukes, R. T. F.; Verhoeven, J. W. Phys. Chem. Chem. Phys. 2002, 4, 1542.
